Abstract

Since their discovery, Wnt signaling molecules have been shown to control key events in embryogenesis, to maintain tissue homeostasis in the adult and, when aberrantly activated, to promote human degenerative diseases and cancer, thus making them a vital area of study. Wnt Signaling: Methods and Protocols examines both biochemical assays and vertebrate and invertebrate model systems to provide a point of reference to current molecular protocols and the diverse model systems employed to study this important signaling pathway. In Volume 1, Pathway Methods and Mammalian Models, assays to measure activation of the diverse Wnt signaling pathways are explored, along with a selection of models and strategies used to study mammalian Wnt/FZD function, from protein-protein interaction and simple cell line models to organoid cultures and mouse models. As a volume in the highly successful Methods in Molecular Biology™ series, chapters contain readily reproducible laboratory protocols, complete with lists of necessary equipment and reagents and the Notes section, which reveals helpful troubleshooting tips. Comprehensive and cutting-edge, Wnt Signaling: Methods and Protocols collects the expertise and knowledge of many leaders in the field to produce this invaluable two-volume resource. .

Abstract

Bacterial infections affect world health today as a leading cause of morbidity and mortality. Pathogenic bacteria routinely command a broad spectrum of niches in the human host, making an understanding of pathogenesis mechanisms crucial to the development of prophylactics and treatment for bacterial diseases. A variety of in vitro methods, in vivo animal model systems and cutting-edge genomics assays have arisen in the effort to study bacterial pathogenesis and identify potential therapeutic targets. In Bacterial Pathogenesis, in-depth methods and state-of-the-art protocols are presented for investigating specific mechanisms of pathogenesis for a wide range of bacteria. This invaluable collection includes protocols to study host-pathogen interactions, animal models of infection, and novel approaches to identifying therapeutic targets designed to control infections. Up-to-date molecular typing methods for Staphylococcus aureus and a new model of streptococcal pharyngitis in non-human primates are also included. Bacterial Pathogenesis will prove an invaluable collection for microbiologists, immunologists, cell biologists and infectious disease clinicians - and indispensable to all science researchers interested in studying pathogenic bacteria and related disease processes.

Abstract

Oxidative Stress in Aging: From Model Systems to Human Diseases discusses the role of free radicals in aging in different animal models, as well as the relevance of free radicals on age-related diseases and pathological conditions in humans (following an introduction section of the basics and theory of free radicals). Human aging is a complex phenomenon - not everyone gets the same diseases and dies from the same cause. Accumulating reports implicate the connection between free radicals and various diseases and age-related pathological conditions. Although the causal relationships have not been established, it is necessary to discuss how free radicals are involved in each situation. In addition, the major interventions trials of antioxidant supplements in age-related disease, cancer and so forth are reviewed and discussed.

Abstract

Our understanding of the apoptotic pathway and its regulation has improved significantly in the last 10 years. However, as our knowledge of the intricate intracellular events increase, we realize the complexity of the regulatory mechanisms that control it. As such, Apoptosis and Cancer describes the performance of contemporary techniques for studying the biology of apoptosis and its role in cancer. This book is a collaboration between academics- and industry-based scientists. The chapters have the technical development characteristic of an academic environment and the standardized system necessary for translating a product to general use, as provided by the industry. Thus, the protocols described within this groundbreaking new volume will aid both an academic laboratory interested in further characterizing the mechanism of apoptosis and an industry laboratory interested on identifying new target molecules or screening for new compounds with potential clinical use. The authors – all leaders in the field – have created a comprehensive text that covers the newest available techniques as well as conventional basic techniques. Apoptosis and Cancer will undoubtedly provide the necessary resources to everyone interested in the area of apoptosis.

Abstract

For researchers today, the avidin-biotin interaction is among the most widely exploited among biomedical scientists. Despite its popularity, avidin-biotin methodology suffers from a lack of clarity associated with popular, low cost, prefabricated kits used by many in the field. In Avidin-Biotin Interactions: Methods and Applications, leaders in avidin-biotin methodology share their knowledge relating to the application of the extremely powerful interaction between the protein avidin or its homologues and the vitamin biotin and some of its homologues. Although avidin-biotin based kits based have been produced for quite some time and remain a widely used methodology, many of the chapters in this text describe situations in which the previous methodology failed due to some factor, and the authors developed new and better ways for the avidin-biotin system to be utilized. With excellent descriptions of laboratory protocols written by expert researchers, this volume is equally perfect for the student or the professional laboratory scientist. Each chapter in Avidin-Biotin Interactions: Methods and Applications contains not only excellent descriptions of protocols for the lab but also the experience of researchers who used specific methodology, found a problem with that methodology in a new setting, and ultimately devised ways to minimize or obviate the limitations of the technology.
